Amplification of 16S rRNA gene sequences to differentiate two highly related bradyrhizobia species PAB

A 16S rRNA gene PCR-based assay was developed aiming at a fast molecular diagnostic method to differentiate the two phylogenetically closely related species Bradyrhizobium japonicum and B. elkanii, isolated from soybean nodules, in order to identify those more competitive and comprising greater nitrogen fixation ability for use in the formulation of commercial inoculants. The assay used was able to discriminate ten reference strains belonging to these two Bradyrhizobium species, as well as to efficiently identify 37 strains isolated from fields cultivated with soybean.

Characterization of plant growth-promoting bacteria inhabiting Vriesea gigantea Gaud. and Tillandsia aeranthos (Loiseleur) L.B. Smith (Bromeliaceae) Biota Neotropica

Microorganisms that live inside and around a plant can supply it with essential substances, such as phytohormones and essential nutrients. The present investigation aimed to isolate and characterize the phyllosphere, the endophytic, and the water tank bacteria associated with Vriesea gigantea and Tillandsia aeranthos. The bacteria were tested for siderophore and indole-3-acetic acid (IAA) production, phosphate solubilization, and presence of the nif H gene. Genetic diversity of the bacterial isolates was evaluated by rep-PCR. Characterization of variants of Bradyrhizobium elkanii and B. japonicum and symbiotic behaviour in soybeans Ciência Rural

Variation in rhizobia strains isn’t a desirable fact based mainly on the possibility of unexpected results on legume inoculation. In this work, we studied the variability on phenotypic characteristics and genetic stability of rhizobia strains recommended for soybean inoculation. Variants with stable colony morphology were obtained from Bradyrhizobium japonicum strain SEMIA 5080 and from B. elkanii SEMIA 5019. Variants from SEMIA 587 obtained by another author were also used. The variants differed on colony characteristics, nodulation capacity, nitrogen fixation efficiency and competitive ability for nodule formation in two soybean varieties (Jacui 7 and IAS 5). Isolation and characterization of two plant growth-promoting bacteria from the rhizoplane of a legume (Lupinus albescens) in sandy soil Rev. Bras. Ciênc. Solo

Two bacterial strains that amplified part of the nifH gene, RP1p and RP2p, belonging to the genus Enterobacter and Serratia, were isolated from the rhizoplane of Lupinus albescens. These bacteria are Gram-negative, rod-shaped, motile, facultative anaerobic, and fast-growing; the colonies reach diameters of 3-4 mm within 24 h of incubation at 28 ºC. The bacteria were also able to grow at temperatures as high as 40 ºC, in the presence of high (2-3 % w/v) NaCl concentrations and pH 4 -10. Strain RP1p was able to utilize 10 of 14 C sources, while RP2p utilized nine. The isolates produced siderophores and indolic compounds, but none of them was able to solubilize phosphate. In the last decades, the use of plant growth-promoting rhizobacteria has become an alternative to improve crop production. Rhizobium leguminosarum biovar trifolii is one of the most promising rhizobacteria and is even used with non-legume plants. This study investigated in vitro the occurrence of plant growth-promoting characteristics in several indigenous R. leguminosarum biovar trifolii isolated from soils in the State of Rio Grande do Sul, Brazil. Promoção de crescimento de arroz por rizóbios em diferentes níveis de adubação nitrogenada Ciência Rural

RESUMO: Embora os rizóbios não apresentem a capacidade de fixar nitrogênio em plantas de arroz, estes podem estimular o crescimento da parte aérea e da raiz, aumentando a absorção de nutrientes, como o nitrogênio, e a produtividade. Com o trabalho, avaliou-se o efeito de rizóbios em promover o crescimento de plantas de arroz em função de doses de nitrogênio (N). Efetuou-se um experimento com vasos contendo solo e plantas da cultivar IRGA 424. As plantas foram inoculadas com oito isolados de rizóbios e adubadas com doses equivalentes a 0, 40, 80 e 160kg N ha-1. Legume species belonging to the genus Lupinus are annual herb plants. The majority of them are indigenous to the Americas. They are known for nitrogen-fixing symbioses with soil bacteria collectively called rhizobia. The aim of this study was to characterize a rhizobium strain isolated from Lupinus albescens using phenotypic, symbiotic and molecular approaches.
